Description

Our Spore to Agar Transfer Kit includes:

MEA and MEAG Plate with Parafilm:

These plates are essential for culturing your spores. MEAG plates are antibacterial, preventing bacterial growth and ensuring a cleaner culture environment.

Scalpel and 10 Blades

For precise cutting and transferring of mycelium.

Sterile cotton Swab

Used to transfer spores from a spore print to the agar plate.

70% alcohol spray bottle

For sanitizing your work surface.

Instructions:

1. Preparing the Plates:
– Use the sterile cotton swab to take spores from a spore print.
– Transfer the spores to a MEAG plate using a ‘Z’ motion. It’s recommended to prepare at least two plates.
– Wear gloves and a mask, and work on a clean, sanitized surface. Spray the surface with 70% alcohol and allow it to sit for 30-45 seconds before use.

2. Observation:
– Monitor the plates for spore colonization. Possible outcomes include:
– **Contamination**: Look for colourful growth such as pink, yellow, black, or green. Note that some contamination starts white and changes colour.
– **Healthy Mycelium**: White growth, but be cautious as some contaminations also start white.

3. Transferring Healthy Mycelium:
– Once you identify healthy mycelium, transfer a healthy piece farthest from any contamination to a new MEA plate.
– This process helps to reduce the genetic pool or number of spore colonies, making it easier to spot hidden contamination.

4. Result:
– If the new plate remains clean after the transfer, you have successfully created a clean agar plate, known as a multi-spore plate. If not repeat Step 3.

Next Steps:
– Use this kit to start your journey towards making Liquid Culture, we have a kit to help you with that too.

This kit provides all the essential tools and guidelines to start your mycology experiments and ensure clean, contamination-free cultures.
